My colleagues and I would like to be able collaboratively edit a cohort that is sync’d to S3. Any way to do that? Are all cohorts are single-member-editable only?

You can ask the cohort owner to hit Share and enter yours and yours colleague’s name.
If this works, you should be able to see the View Only turn to Edit Access for that cohort.
